轉(zhuǎn)化細(xì)胞存活測(cè)試
閱讀:92 發(fā)布時(shí)間:2017-11-17(1)計(jì)算轉(zhuǎn)化細(xì)胞數(shù)目可用血球計(jì)數(shù)盤或是Coultercounter粒子計(jì)數(shù)器自動(dòng)計(jì)數(shù)。
(2)血球計(jì)數(shù)盤一般有二個(gè)chambers,每個(gè)chamber中細(xì)刻9個(gè)1mm2大正方形,其中4個(gè)角落之正方形再細(xì)刻16個(gè)小格,深度均為0.1mm。當(dāng)chamber上方蓋上蓋玻片后,每個(gè)大正方形之體積為1mm2×0.1mm=1.0x10-4ml。使用時(shí),計(jì)數(shù)每個(gè)大正方形內(nèi)之細(xì)胞數(shù)目,乘以稀釋倍數(shù),再乘以104,即為每ml中之細(xì)胞數(shù)目。
(3)存活測(cè)試之步驟為dyeexclusion,利用染料會(huì)滲入死細(xì)胞中而呈色,而活細(xì)胞因細(xì)胞膜完整,染料無法滲入而不會(huì)呈色。一般使用藍(lán)色之trypan blue染料,如果細(xì)胞不易吸收trypan blue,則用紅色之Erythrosin bluish。
2、材料:
0.4%w/v trypan blue(GibcoBRL15250-061);Erythosin bluish stain;取0.1gram Erythrosin bluish(SigmaE-9259)及0.05gram preservative methyl paraben(SigmaH-3647)溶于100mlCa++/Mg++freesaline;血球計(jì)數(shù)盤及蓋玻片(Hemocytometerandcoverslip);計(jì)數(shù)器(counter);低倍倒立顯微鏡;粒子計(jì)數(shù)器(Coultercounter,CoulterElectronics)。
3、步驟:
(1)取50μl細(xì)胞懸浮液與50μl trypan blue(orErythrosinbluish)等體積混合均勻于1.5ml小離心管中。
(2)取少許混合液(約15μl)自血球計(jì)數(shù)盤chamber上方凹槽加入,蓋上蓋玻片,于100倍倒立顯微鏡下觀察,活細(xì)胞不染色,死細(xì)胞則為藍(lán)色(或紅色-Erythrosin bluish)。
(3)計(jì)數(shù)四個(gè)大方格之細(xì)胞總數(shù),再除4,乘以稀釋倍數(shù)(至少乘以2,因與trypanblue等體積混合),zui后乘以104,即為每ml中細(xì)胞懸浮液之細(xì)胞數(shù)。若細(xì)胞位于線上,只計(jì)上線與右線之細(xì)胞(或計(jì)下線與左線之細(xì)胞)。
注:4大格細(xì)胞總數(shù)×稀釋倍數(shù)×104/4=細(xì)胞數(shù)/ml;每一大格的體積=0.1cm×0.1cm×0.01cm=10-4ml
4、范例:
T75 monolayer culture制成10ml細(xì)胞懸浮液,取0.1ml溶液與0.1ml trypan blue混合均勻于試管中,取少許混合液加入血球計(jì)數(shù)盤,計(jì)數(shù)四大方格內(nèi)之細(xì)胞數(shù)目。
活轉(zhuǎn)化細(xì)胞數(shù)/方格:55,62,49,59;死細(xì)胞數(shù)/方格:5,3,4,6;細(xì)胞總數(shù)=243
平均細(xì)胞數(shù)/方格=60.75;稀釋倍數(shù)=2
細(xì)胞數(shù)/ml:60.75×104×2(稀釋倍數(shù))=1.22×106
細(xì)胞數(shù)/flask(10ml):1.22×106×10ml=12.2×106
存活率:225/243﹦92.6%
